Introduction
Purpose/Methods
In the salt experiment, the first step was to create the stock solutions. Test tube number one was prepared with 8 mL of 0.5% starch solution, 1 mL pH7 solution, 6 mL water, and 1 mL of amylase. Test tube number two was prepared with 8 mL of 0.5% starch solution, 1 mL pH7 solution, 3 mL of salt concentration, 3 mL water, and 1 mL of amylase. Test tube number three was prepared with 8 mL of 0.5% starch solution, 1 mL pH7 solution, 6 mL of salt concentration, 0 mL water, and 1 mL of amylase. Test tube number four was prepared with 8 mL of 0.5% starch solution, 1 mL pH7 solution, 0 mL of salt concentration, 6 mL water, and 0 mL of amylase. The wavelength was 605. After the stock was prepared, 1 mL of the solution from each of the tubes was added to another tube containing 2 mL of water and 3 drops of potassium iodine every 3 minutes for 18 minutes.
Results
Figure 5.1 Salt’s Effect on the Enzyme Amylase. This graph shows the effect of salt on the enzyme amylase. The control was the variable without enzyme. Its data remained constant at 2.5 absorbance. The other variables tested, however, did not remain constant. Their data all started at 2.5 absorbance, then decreased over time. Each salt concentration produced very similar amylase activity at each interval.
Figure 6.1 pH’s Effect on the Enzyme Amylase. This graph shows the effect of pH on the enzyme amylase. The control was the variable without enzyme. Its data remained constant at 2.5. The pH 10 sample also remained at 2.5 absorbance throughout the experiment with no variation. The pH 4 sample remained constant at 2.5 absorbance until the 12 minute time interval. At 12 minutes, the amylase activity decreased. At pH 4’s 15 minute time interval, there was a .1 increase in enzyme activity, but then it decreased again at the concluding 18 minute interval. The pH 8 sample showed more decrease in enzyme activity than no enzyme, pH 10, and pH 4. The pH 8 showed constant decrease throughout the experiment with a steep decline between the 12 and 15 minute time interval. The pH 6 sample showed the largest amount of decrease in the experiment. The absorbance reading shows how much particle the sample contains.
The substrate in the experiment is starch. Starch combines with the enzyme amylase to form the enzyme substrate complex. Maltose is the product. The absorbance reading shows how much particle the sample contains. The less absorbance in a reading, the less starch present. As the amount of substrate decreases the enzyme is working.
